This dissertation evaluates the effectiveness of polyols (xylitol, sorbitol, erythritol) and the probiotic Lactobacillus salivarius HM-6 Paradens in inhibiting Streptococcus mutans and reducing caries progression in children. The in vitro study showed that erythritol most effectively inhibited S. mutans biofilm formation, especially on glass ionomer surfaces, while xylitol increased total biomass. The second study, involving 127 children, found that 2- week probiotic supplementation significantly reduced the increase in dentin and cavitated caries (ICDAS 3-6) after 12 months. No effect was observed on early lesions (ICDAS 1-2). However, no reduction in dental plaque was recorded in the probiotic group. Findings suggest that replacing sucrose with erythritol and using glass-ionomers may improve caries prevention and treatment. Probiotics with L. salivarius may support remineralization of existing lesions but not their initiation.
