The aim of the work was to characterise perivascular inflammation in the early stages of atherosclerosis and to determine how stimulation of the Mas receptor by AVE0991 affects the inflammatory mechanisms of atherosclerosis. Results showed that AVE0991 has anti-inflammatory effects on perivascular adipose tissue by reducing leukocytes content (especially macrophages, T cells, NK cells and dendritic cells) at early stage atherosclerosis. Agonism of Mas receptor by AVE0991 results in reduction of atherosclerotic plaque area and improved stability by reduction of macrophages content in atherosclerotic plaque. Anti-inflammatory effects of AVE0991 are primarily associated with decreased gene expression of pro-inflammatory and chemotactic agents in perivascular adipose tissue. Mechanism of action of AVE0991 is also associated with a reduced polarization to M1 macrophages as well as by decreased chemotaxis of pro-inflammatory monocytes.
